Wineglass bay is a great trip to make.
Belinda and I have done this as a day trip, however, it is a solid drive. You would need to leave Hobart by 8.30 am, after having breakfast at Jampacked (Book) which opens at 7am, then drive the 3hrs, do the walk, then have lunch at the cafe and head back or of course stay over night if time permits.
I would suggest that to really enjoy the area you need at least 1 night staying there. Do the walk, and take the boat trip the next day. This will give you a much better experience without rushing it all.
